It depend on who will answer the phone number written on your reference
letter, either the one on the letter head, or on the letter itself if
any.

Usually the will ask for Human Resources Department first asking wether
they really wrote the reference letter and the information about you is
absolutely correct. If the answer still leaving any hesitation then
they will contact your manager/boss via HR.
