If you've never been, I'd assume Kyoto would hold more for you to
explore. Many museums, temples, shrines, palaces, famous gardens, etc.

Dig through Osaka as you would the others, for a day or more. Hiroshima
is kind of a shlep from Kyoto.

We always fly in to Osaka and spend the "turn-around" night plus
another full day and night. This to get our sleep schedule under
control. In the process we've gotten to where we really like the
mid-town area and look forward to it each time. Nightlife is plentiful
as Unke notes, and there's lots of great food.

But comparitively, it's sight-seeing deprived as you'll find as you
flip through the tourist books.
