Technically possible, the problem will be finding a boat. There are
boats leaving at 7am from Siem Reap to Phnom Penh, once in PP you'd have
to find a boat to continue the journey. Then there are 3-5 day tours
from Siem Reap to Saigon (bookable in Siem Reap).

I was twice in Siem Reap and both times I just hired a local. The second
time I hired the taxi driver who picked me up at the airport for $25/day
(car + driver), which is cheap in my opinion.

As I wrote it is possible to book tours in Siem Reap.
