As an Eastern Shore native I'll try to help. You can drive to Easton on Rt.
50 and take the Easton bypass (MD Rt 322) to St. Michaels Road (Md Rt. 33)
and drive and explore all the way to Tilghman Island (about 24 miles from
Easton.) When you finish exploring there you can overnight in Easton
(Holiday Inn Express and Day's Inn on Rt. 50 and the Tidewater Inn in
downtown Easton.) You can also explore Oxford (Rt. 333 off the Easton
Bypass). This town is fairly small (800 population) but has many old
buildings (including the first Naval Academy) and the oldest ferry in the
US. (unfortunately it is closed currently for reconstruction of the Oxford
ferry dock otherwise you could take the ferry from just off Rt 33 to
Oxford.) The Robert Morris Inn (at the end of Md 333 in Oxford) serves one
of the best crab cake dinners around. I like the Morris (broiled not
fried). Sunday there is a special brunch but they close at 2:30 PM so get
there early (I think they go full days beginning of May).

Other "quaint places to explore are Chestertown, Centreville and the areas
between. (Use MD 213 off of US 301 for the "scenic drive").

Stay away from Kent Island for exploring (just like being in the city) but
Queenstown (just North of US Rt. 50 split on 301) is another place to
explore.
