I used to work at a retail store. Clerks sometimes don't like to
accept $20s for small purchases because it's easy to run out of small
bills if you do that too often, because you're giving back a lot of
change.

For example, if the clerk is running low on $1 bills, and a customer
wants to give them a $20 to pay for a pack of gum, the clerk may ask
if the customer can pay with a smaller bill.

Also, the concern about counterfeit money that folks have mentioned
can also be an issue.
