Midtown is much more convenient than downtown for most travelers.

Little Italy, the South Street Seaport, and Rockefeller Center are all
total tourist traps and not worth your time. Generally avoid
restaurants in those areas, and in Times Square and the Theater
District.

Don't go to La Marqueta in Spanish Harlem (which is featured in some
older guidebooks) - once a thriving Latin market, there's almost
nothing left there.

The Staten Island Ferry is worth taking for the views, but when you
get to the other side get on the return ferry immediately and don't
hang around on Staten Island, which has nothing of interest at all.
