I've used AMTRAK for both the New York-Buffalo trip (2006)
and Washington-New York last month.

If you choose a morning departure the New York-Buffalo trip
is a pleasant one of about 8 hours with quite interesting
scenery as it follows the Hudson north then turns left
towards Buffalo. I alighted at Depew, which is in them
middle of nowhere, but my hotel had a shuttle from there.
I'm not sure how far the downtown station is from hotels.

The Washington-New York trip was also interesting from the
train; it takes about 3 hours depending on whether you get
an express or a regional. The price also varies depending on
your selection. You may not save a lot of money - sometimes
the specials for flights available on Expedia or Priceline
can be cheaper than the train. However, don't forget the
cost of getting to and from the airport at each end.

I prefer trains to buses; much more pleasant, able to walk
around and usually quicker.

Consider renting a car. We picked one up in Buffalo for $18
per day (Avis, via Priceline) and wandered around up-state
New York, New England, Quebec and Ontario in a loop over 12
days.
